What it is

CJC-1295 no DAC — also called Modified GRF (1-29) — is a synthetic 29-amino-acid analog of growth-hormone-releasing hormone. Without the drug-affinity complex (DAC) of the long-acting form, it clears the plasma within minutes, producing a short, pulsatile signal at the GHRH receptor.

It is used in the laboratory as a short-acting reference compound in somatotropic-axis and pituitary-signaling research, frequently studied alongside a selective ghrelin-receptor agonist. Supplied strictly for research use.

    Three maleimido derivatives of hGRF(1-29) were synthesized and conjugated ex vivo to human serum albumin; the conjugates were assessed in vitro for stability against dipeptidylpeptidase-IV and for activity in a growth hormone secretion assay in cultured rat anterior pituitary cells, and the derivatives were administered subcutaneously to normal male Sprague Dawley rats with plasma growth hormone measured. CJC-1295, described in the paper as a tetrasubstituted hGRF(1-29) bearing an N-epsilon-3-maleimidopropionamide lysine at the C terminus, gave the largest 2-hour growth hormone area under the curve and was selected for pharmacokinetic evaluation, in which it was detected in plasma beyond 72 hours.

The frequency and magnitude of growth hormone secretory pulses were reported as unaltered, while basal (trough) growth hormone levels, mean growth hormone levels and IGF-I levels were higher after administration; IGF-I changes did not correlate with any measured parameter of growth hormone secretion.

    An unknown pharmaceutical preparation submitted in 2009 by Norwegian police and customs authorities was analysed by liquid chromatography-high resolution tandem mass spectrometry and found to contain a 29-amino-acid peptide with a C-terminal amide function; a sequence was proposed from the mass spectrometric data and reported as consistent with the peptide marketed under the name CJC-1295. No cell, tissue or animal experiment was performed. The paper states that CJC-1295 is a releasing factor for growth hormone and is therefore considered a Prohibited Substance under Section S2 of the WADA Prohibited List.
